What is IDRA21?
IDRA-21 is a positive allosteric modulator of AMPA receptor and a benzothiadiazine derivative. It is a chiral molecule with (+)-IDRA-21 as the active form.
IDRA-21 has shown nootropic effects in animal studies, significantly improving learning and memory. In reversing cognitive deficits caused by alprazolam or scopolamine, it is approximately 10-30 times more effective than Aniracetam, and produces a sustained effect that lasts up to 48 hours after a single administration. The mechanism of this action is thought to be by promoting the induction of long-term potentiation between synapses in the brain.
IDRA-21 may not produce neurotoxicity under normal conditions, although it may aggravate neuronal damage after a stroke or seizure after a systemic ischemia.
Compared with AMPA receptor enhancers derived from ampakin or benzoylpiperidine, IDRA-21 is more effective than CX-516, but not as effective as CX-546. New benzothiadiazine derivatives with greatly improved potency compared with IDRA-21 have been developed, but these have not been studied to the same degree. Benzoylpiperidine and benzoylpyrrolidine CX series of drugs are more subject to clinical development. Favor, most probably due to the more favorable toxicity profile at higher doses.
